Introduction. Ceftriaxone is an effective cephalosporin antibiotic, but its use can lead to undesirable effects. Directed transport of drugsreducesthese effects, the therapeutic dose of the drug and the frequency of administration. Aim. To develop a ceftriaxone delivery system using erythrocyte vectors for non-obstructive pyelonephritis to selectively affect the organ in order to increase its efficiency and reduce toxicity
Materials and methods. Studies conducted on rats, non-obstructive pyelonephritiswas modeled by intraperitoneal administration of arachidonic acid. The state of the excretory function of the kidneys was judged by the concentration of urea and creatinine in the blood. To incorporate ceftriaxone into the erythrocyte stroma, the method of hypoosmotic hemolysiswas used. Ceftriaxone was quantitated by HPLC.
Results. It was established that five-fold administration of ceftriaxone to animals with pyelonephritis normalized body temperature for 10-12 days, excretory kidney function for 12-14 days.The number of leukocytes wasincreased, the number of lymphocytes decreased, the number of neutrophils did not change, decreased, but the imbalance of enzyme activity in the kidneys did not normalize.Asingle administration of erythrocyte form of ceftriaxone to ratswith pyelonephritis normalized body temperature for 5-7 days, excretory kidney function – for 5-8 days, respectively, on the 10th day full normalization of the number of leukocytes, lymphocytes, neutrophilsin the blood, activity was noted enzymes in the kidney.
Conclusion.Thus, a ceftriaxone delivery system was developed using erythrocyte carriers for non-obstructive pyelonephritis.
